چکیده
Aim: The present study was aimed at polymerase chain reaction (PCR) amplification and cloning of NAD-dependent betahydroxybutyryl coenzyme A dehydrogenase (BHBD) gene of Clostridium chauvoei. Materials and Methods: C. chauvoei was cultured and confirmed by 16-23S rDNA spacer region primers. The primers for nad-bhbd gene of C. chauvoei were designed to aid in cloning into pRham-N-His SUMO-Kan vector, and nad-bhbd gene was amplified by PCR. The amplified nad-bhbd gene was purified and cloned into pRham-N-His SUMO-Kan expression vector. The recombinant plasmid was transformed into E. cloni 10 G cells and the clone was confirmed by colony PCR using the pRham-SUMO-NAD-For and pRham-SUMO-NAD-Rev primers and also by sequencing. Results: PCR amplification of nad-bhbd gene yielded a product length of 844 base pairs which was cloned into pRham-NHis SUMO-Kan vector followed by transformation into E. cloni 10G chemically competent cells. The recombinant clones were characterized by colony PCR, sequencing, followed by basic local alignment search tool (BLAST) analysis to confirm the insert. Conclusions: Immunogenic protein NADdependent BHBD of C. chauvoei was cloned and the recombinant clones were confirmed by colony PCR and sequencing analysis.

Cloning and sequence analysis of hyaluronoglucosaminidase (nagH) gene of Clostridium chauvoei
AIM Blackleg disease is caused by Clostridium chauvoei in ruminants. Although virulence factors such as C. chauvoei toxin A, sialidase, and flagellin are well characterized, hyaluronidases of C. chauvoei are not characterized. The present study was aimed at cloning and sequence analysis of hyaluronoglucosaminidase (nagH) gene of C. chauvoei. MATERIALS AND METHODS C. chauvoei strain ATCC 10092...

Background: Clostridium chauvoei is the etiological agent of blackleg as an endogenous infection in cattle. Flagella have been known to play a critical role in the protective immunity of animals to clostridial infections. C.chauvoei has two copies of fliC gene, namely fliA and fliB. OBJECTIVES: The aim of this study was the determination and nucleotide sequence analysis of both copies of fliC ...
متن کاملFusion of Clostridium perfringens type D and B epsilon and beta toxin genes and it’s cloning in E. coli
Designing and producing a proper fusion construction is the most important problem of producing large quantities of a properly folded functional protein. This construction should have all necessary components of a real gene. A good designed fusion gene construction could be cloned into a good and suitable host. Clostridium perfringens is an important pathogen of humans and livestock and produce...
متن کاملIsolation of a new butanol-producing Clostridium strain: high level of hemicellulosic activity and structure of solventogenesis genes of a new Clostridium saccharobutylicum isolate.
New isolates of solventogenic bacteria exhibited high hemicellulolytic activity. They produced butanol and acetone with high selectivity for butanol (about 80% of butanol from the total solvent yield). Their 16S rDNA sequence was 99% identical to that of Clostridium saccharobutylicum. The genes responsible for the last steps of solventogenesis and encoding crotonase, butyryl-CoA dehydrogenase, ...
متن کاملCloning of Clostridium perfringens iota toxin gene in Escherichia coli
Iota toxin is produced by Clostridium perfringens type E. This toxin causes antibiotic-associated enterotoxemia in lambs and calves. Iota toxin is a binary toxin that has two components including Ia (the enzyme component) and Ib (the binding component). Ib binds to the surface receptor of target cells and translocate Ia into the cytosol of cells. The aim of this study was to clone toxigenic epi...
